Terbinafine is an allylamine antifungal agent with potent activity against a wide range of fungi. It works by inhibiting the enzyme squalene epoxidase, which is essential for fungal ergosterol biosynthesis. Ergosterol is a vital component of the fungal cell membrane. Inhibition of this pathway leads to ergosterol deficiency and intracellular accumulation of squalene, resulting in disruption of cell membrane integrity and fungal cell death.
Terbinafine is fungicidal against dermatophytes, molds, and certain dimorphic fungi, and may be fungicidal or fungistatic against yeasts depending on the species. Topical terbinafine has demonstrated high efficacy in infections of the skin caused by Trichophyton, Microsporum, Epidermophyton, and Candida species, as well as Malassezia furfur responsible for pityriasis versicolor.
