Glipita M XR Tablet 50 mg+500 mg is a fixed-dose combination of Sitagliptin and Metformin Hydrochloride (Extended Release), designed for the effective management of type 2 diabetes mellitus in adults. This combination therapy targets multiple pathophysiological defects of diabetes, helping achieve better and sustained glycemic control when lifestyle modification alone or monotherapy is inadequate.
The extended-release formulation of metformin ensures gradual drug release, improving gastrointestinal tolerability and allowing convenient once-daily dosing.

DPP-4 Inhibitor + Biguanide (Antidiabetic Combination)
Sitagliptin is a selective dipeptidyl peptidase-4 (DPP-4) inhibitor. It increases endogenous incretin hormone levels (GLP-1 and GIP), which enhance glucose-dependent insulin secretion and suppress glucagon release from the pancreas. This leads to reduced hepatic glucose production and improved postprandial and fasting blood glucose control.
Metformin Hydrochloride primarily reduces hepatic glucose output and improves peripheral insulin sensitivity by increasing glucose uptake and utilization in muscle tissues. It also decreases intestinal glucose absorption. The extended-release formulation provides sustained plasma levels and reduces common gastrointestinal side effects associated with immediate-release metformin.
The complementary mechanisms of action of Sitagliptin and Metformin result in effective glucose control with a low risk of hypoglycemia.
Glipita M XR Tablet is indicated for:
Treatment of type 2 diabetes mellitus in adults
Patients inadequately controlled on metformin or sitagliptin alone
Use as add-on therapy along with diet and exercise
The usual recommended dose is one tablet once daily, preferably taken with the evening meal. Tablets should be swallowed whole and not crushed, chewed, or split. Dose adjustments should be individualized based on glycemic response, tolerability, and renal function. Treatment should be initiated and supervised by a registered physician.
