A-Clox Capsule 500 mg is indicated for the treatment of infections caused by cloxacillin-sensitive organisms, including:
Skin and soft tissue infections such as boils, abscesses, cellulitis, and wound infections
Bone and joint infections including osteomyelitis
Respiratory tract infections
Septicemia caused by susceptible staphylococci
Endocarditis
Post-operative and post-traumatic infections
It is particularly useful in infections where resistance to ordinary penicillins is suspected or confirmed.
Penicillinase-resistant penicillins (β-lactam antibiotics)
Cloxacillin Sodium exerts a bactericidal action by inhibiting bacterial cell wall synthesis. It binds to penicillin-binding proteins, interfering with the final stage of peptidoglycan synthesis, which is essential for bacterial cell wall integrity. This results in cell lysis and death of susceptible bacteria.
Cloxacillin is stable against staphylococcal penicillinase enzymes, which allows it to remain effective against penicillin-resistant strains of Staphylococcus aureus. After oral administration, cloxacillin is moderately absorbed from the gastrointestinal tract and is widely distributed in body tissues and fluids. It is primarily eliminated via renal excretion.
A-Clox Capsule 500 mg inhibits bacterial cell wall synthesis, leading to weakening and rupture of the bacterial cell wall. Its resistance to β-lactamase enzymes ensures sustained antibacterial activity against penicillinase-producing staphylococci.
Dosage depends on the type and severity of infection:
Adults: 250–500 mg every 6 hours
Severe infections: Higher doses may be prescribed under medical supervision
A-Clox Capsule should be taken on an empty stomach, preferably 1 hour before or 2 hours after meals, to ensure optimal absorption. Treatment duration should be completed as prescribed to prevent resistance.
Cloxacillin may reduce the effectiveness of oral contraceptives. Concomitant use with bacteriostatic antibiotics such as tetracyclines may reduce its antibacterial activity. Caution is advised when used with anticoagulants.
A-Clox Capsule 500 mg is contraindicated in patients with:

History of severe allergic reactions to β-lactam antibiotics
