Indications

Acmegra Tablet 50 mg is indicated for the treatment of:

• Erectile dysfunction in adult males

• Difficulty in achieving or maintaining penile erection adequate for sexual intercourse

Sildenafil works only when sexual stimulation is present and does not increase sexual desire.

Therapeutic Class

Pharmacology

Sildenafil Citrate exerts its therapeutic effect by selectively inhibiting phosphodiesterase type 5, the enzyme responsible for the degradation of cyclic guanosine monophosphate (cGMP). During sexual arousal, nitric oxide is released in the corpus cavernosum, leading to increased cGMP levels. Elevated cGMP causes relaxation of smooth muscle and enhanced blood flow into the penile tissue. By preventing cGMP breakdown, Sildenafil prolongs and enhances the erectile response. In the absence of sexual stimulation, Sildenafil has no pharmacological effect.

Dosage and Administration

The usual recommended starting dose of Acmegra Tablet is 50 mg, taken approximately 30 minutes to 1 hour before anticipated sexual activity. Based on individual response and tolerability, the dose may be adjusted to a maximum of 100 mg or reduced to 25 mg.

• Maximum dosing frequency is once daily

• Tablet should be swallowed whole with water

• May be taken with or without food; however, a high-fat meal may delay onset of action

Dose adjustments may be required in elderly patients and in those with hepatic or severe renal impairment.

Drug Interactions

Acmegra Tablet 50 mg should not be used in combination with nitrates or nitric oxide donors due to the risk of severe hypotension.
