A-Clox Suspension 125 mg/5 ml is indicated for the treatment of mild to moderate infections caused by cloxacillin-sensitive organisms, including:
Skin and soft tissue infections such as impetigo, boils, abscesses, and infected wounds
Respiratory tract infections
Bone and joint infections, including osteomyelitis
Ear, nose, and throat infections
Septicemia due to susceptible staphylococci
Post-operative and post-traumatic infections
It is particularly effective in infections where resistance to ordinary penicillins is suspected.
Penicillinase-resistant penicillins (β-lactam antibiotics)
Cloxacillin Sodium is a semisynthetic β-lactam antibiotic that exhibits bactericidal activity by inhibiting bacterial cell wall synthesis. It binds to penicillin-binding proteins, disrupting the formation of peptidoglycan, an essential component of the bacterial cell wall. This leads to weakening of the cell wall and eventual bacterial lysis.
Cloxacillin is stable against staphylococcal penicillinase enzymes, allowing it to remain effective against penicillin-resistant strains of Staphylococcus aureus. After oral administration, cloxacillin is absorbed from the gastrointestinal tract and distributed widely throughout body tissues and fluids. The drug is primarily excreted through the kidneys.
A-Clox Suspension interferes with bacterial cell wall synthesis by inhibiting peptidoglycan cross-linking. Its resistance to β-lactamase enzymes ensures sustained antibacterial activity against penicillinase-producing staphylococci.
Dosage depends on age, body weight, and severity of infection:
Children: As prescribed by a physician, usually in divided doses every 6 hours
Adults (if required): Dose adjusted according to clinical need
The suspension should be taken on an empty stomach, preferably 1 hour before or 2 hours after meals, to maximize absorption. Shake the bottle well before each use and complete the full course of therapy as directed.
Concomitant use with bacteriostatic antibiotics such as tetracyclines may reduce the antibacterial effect of cloxacillin. Cloxacillin may also reduce the efficacy of oral contraceptives. Caution is advised when used with anticoagulants.

Most adverse effects are mild and reversible upon discontinuation.
A-Clox Suspension may be used during pregnancy and lactation under medical supervision, as cloxacillin is considered relatively safe in these populations.
Store in a cool, dry place below 30°C. After reconstitution, store as directed on the label and use within the recommended period. Protect from light and keep out of reach of children.
